Time Essay Part I:The Tao Of DaniTime is an illusion because there is no real truth to it. It has never been proven to exist. It was made up by the Romans or the Babylonians thousands of years ago so farmers could meet on a hill top at 1 in the afternoon because one farmers wife had a hair appointment at noon. Human beings have a born inhibition to have things simpler. Human beings also have to have explanations and reasoning. Time was born out of this. The grand old theory that time is a river. The whole river exists at once: the mouth (future), the main body (present), and the source (past). The only truth we have to time is the wear and tear on our bodies as time (sic) goes on. Technically, there is no now and there is no future because now was over a nano-second ago and the future is uncertain (but the end is always near). Which is plain truth because the end is nearer and nearer but youll never know it. Flowing, clumping,sparking, rolling across nothing to make something. A few years later, the universe is formed. Things evolve and change and yet everything stays the same. Cells become smart and grow nuclei, which form unicellular organisms, everything feeds off everything else. Things die off, some things live, animals come to be, and all of a sudden you have humans that are just as much fish as they are bacteria as they are stardust. Everything depends on everything else. The universe is me because I am human, thusforth made up of everything and doing my part to keep the world going rather I like it or not. I am the universe because I can almost control what goes on. I am also the universe because I am part of it, we move together, we live together, we love together. This I am the universe can also be explained in a sense of which that humans make themselves gods. We dont hug trees, we crap on them. We dont love animals, we bu tcher them. But we have to do this to survive. Then we ponder meaninglessly at the problems of overpopulation and natural disasters. You are part of natural disasters, like it or not. The distinction between been, being, and becoming is artificial and arbitrary because if time is irrelevant, then you could never have been, or you could never become, you just are. Like the river, you are everything all at once. At any given instance, you bring all your conscious and subconscious knowledge to the table for discussion. All of your past experiences, dreams, realizations, mishaps, good hair days, etc. are with you forever as you go along. You never leave anything behind, therefore you could not possibly have ever been. (This also opens up a discussion about how every nanosecond of your life exists at once; the theory of dimensions.) Everything you are to become is already chosen for you by something called destiny, you just arent aware of it yet. This is also due to post experiences, etc. The distinction between the three aforementioned is that there is no distinction because of the artificial and arbitrary sense. (If something isnt there, it doesnt exist.) Humans make things up to make life easier. They made the theory of been, being, and becoming up to make it easier to convey thoughts. Humans learned to speak to improve their life styles. All the other organisms have been doing just since the big bang, without a spoken or written language. Time is related to the unity of all things in the fact that over time the universe has evolved and change to become what it is today. In fact, the universe will still evolve and change until it reaches its limit and begins devolving. The universe is me at this moment because I am here and know that everything around me is due to me. This is just as I am due to everything around me. This is due to time. I know at this moment, because I am here at this moment, aware at this moment that I am the universe and the universe is me. Dont ponder the statements, just believe. The second you start to ask questions is the second you dont understand. The second you understand is the second time has chosen for you to understand, and you will ask no questions. The universe can very vaguely be explained, as the human psyche can very vaguely be explained. In the time it takes for you to understand these concepts, another three thousand people have died, another six women in the Bronx have been raped. Time is all relative because it effects you and yet it doesnt. Maybe one of those women who was just raped died, and maybe she was the one to discover the cure to a fatal disease you will have in later life. So now you will die at 60 instead of 127. Yet, time says that this will happen, but you have no clue of this until you are on your deathbed, and maybe not even then. The universe is everything around you. Everything that changes and evolves over time. Yet, time doesnt exist. So the simplest answer is that things happen because they happen. Thats just how the universe works. Time is related to the distinction between been, being and becoming because if there is no time, then there is no been, being or becoming. There is only a NOW. As inETERNITY IS NOW and it is because when the hell else are you going to live? All that you can see, hear, feel, and taste, all that you know is in the present, so how could you live any other way? You cant. You just have now, so get with it and realize that there will never be a past. Its dreams, bits and pieces of memory. Whats done is done. Jim Morrison is never coming back (as Jim Morrison, anyway). The future cannot be told by a chick in a golden turban with gaudy jewelry on. Your future is set for you, true, but you wont know until you get there, and when you get there, it will be the past before you know it. The only thing you can hold on to is now. So hold on to it because its all you have to hold on to and you cant even do that. The unity of all things is related to been, being and becoming because everything has its place in the universe you see (and dont see) right now. Everything had to come from somewhere, just as we have discussed in previous paragraphs. The details of this are obscured by clouds, so to speak, in the fact that you cannot see all the incarnations of everything at once. Unless you posses the sixth sense (or your brain works in more places than a normal human brain) you cannot see all the incarnations of this paper at once. You never will either, due to time moving on and on and the universe going with it. I look up, it looks like the buildings are burning, but its just the sun setting; the solar system calling an end to another business day. Eternally circling signaling the rhythmic clicking on and off of computers. The pulse of the American machine. The pulse that draws death dancing out of anonymous side streets you know the ones that always get dumped on and never get plowed. It draws death dancing out of little countries with funny languages where the ground is getting harder and it was not that soft before (Ani DiFranco, Not So Soft). The previous quote proves a point that I wish to make- universe is one because everything is run (at least to us Earthlings) by a sun and a moon giving us light and dark. The universe goes on and on, circling around for all eternity and we are only aware of a moment at a time. A pulse beating in anothers absence. In deep thought, the universe clicks into place as being everywhere at the same time where in fact the universe just is. Time not existing is a concept that I would like to bring up at a formal dinner somewhere, so as to confuse the people around me. My knowledge of this concept also makes a good conversation with Allan Stagg. Spiritually, it does nothing for me. Being raised in a western religious house, I never believed in one all -powerful being because thought it was stupid. I figured out at a young age that it really doesnt matter what you do in your life. Your life is controlled by something called destiny, and nothing can change that. Time fits into this because it seemed to me that some guy living for years upon years in the clouds was bull*censored*. I came to the conclusion that everything they told me was made up by old men with small *censored*s in a bar with no lives. Hell, the Bible has been translated, lost, and re-written so many times and youre going to tell me that that is the real story. That the Bible is Gods word?! How the hell do you know that? Theres no proof. Wouldnt it be hilar ious if all this time youve been believing bull*censored*?To make a better life for myself using the second concept is simple: realizing that there was a me decade for a reason, and how all the Aberzombies came to be. I am the universe and the universe is me because, like I have money, and everyone should, like, bow down to me (typical Aberzombie). Seriously, I now understand the circle of life in a more philosophical, holistic view, which makes me a more well-rounded person. I think that everything is made up of everything, and I always have thought this. Maybe its because I learned it off Sesame Street. I dont know. Alls I know is that the universe is an illusion as I am an illusion and whatever anybody else says is bull*censored*. There is no such thing as California because it doesnt exist yet. There are lines on a map and taxes being paid, signs and roads, even an ocean for it to fall into in the near future, but California does not exist. The six degrees of Kevin Bacon exists as the six degrees of me exists- in theory. The universe was created with a bang and it goes on from there. The theories we not developed until people began to be inquisitive, when maybe it would have been simpler to not have been inquisitive at all. But we, as a human kind, have gotten ourselves here and now, and now we have to live with our past lives decisions. I have never been, I will never become, I just am. Im here, now, lets play the game of life and get it over with. Again, this is another fun conversational piece at a formal dinner. These three concepts that do not exist, effect my life in the fact that I now know everything exists at this moment. Ive always had the theory that for every nanosecond of your life, there is a different dimension and a million plus dimensions. The futures uncertain and the end is always near (Jim Morrison, Roadhouse Blues). The future will never be clear and once it is, it is the past. The dead are always with us, and will never leave us. They are a part of us as long as we are all here. Their spirit stays with the living because of what they left behind- impressions on our minds. I will always be in the now, and so will everything else that popped up around here. Living in the past is something of a bad thing because you can never change your past- its just there. Many people live in the past and play t he what if game. Many of these people are seek and do not seek professional attention for their problems, but should. These people are never happy and probably never will be truly happy. So why be like them and live in the past, when you can look to your future and possibly have the chance to be happy? Living in the now is what gives you the keys to life. Not just flying by the seat of your pants, but thinking things out, and having the knowledge to fly by the seat of your pants and not get, into a bad situation voluntarily. And if you do get into a negative situation, having the knowledge to get out of it and it becomes wisdom. Being is something we all must deal with and in the process we must make ourown paths. There are many paths you can go by on any given day. The path you choose isyour choice. If I have learned one thing over the years it is that if it is not you who isdoing the thinking, then you are not living. You must think for yourself at all time in orderto live a fulfilling and happy life. You can not let other people drive your car if you aredriving it. Let them be uncomfortable as long as you are not. It is your life, you can dowhat you want with it. You only get one chance at this game (that you know of, unlessyou follow an ism then in which case you have many chances), better to make the bestof it than to throw your life away. Time wasted is never regained. That was what waswritten under the clock in the band room at a school I used to attend. If you waste yourtime with petty things, you will be reduced to having only petty things. Decide what youreally want and go after it. If you do not k now what you want then go after a lot of things,but do not burn yourself out. Go after what your heart desires and listen to your heart. Live your life like you are always being followed by a camera, and remember that you arethe director and the star- nobody likes somebody whos full of themselves. Only shallowpeople will surround you if you are like that. And remember to never play the what ifgame. The past is gone and done with. Yeah, *censored*s going to happen. Milk will spill. Getover it and get on with your life because you only have a short time here to prove yourselfto the audience, and if thats what you seek, so be it. Remember, everything must comefrom within, and a movie without a good, action-packed plot sucks. The past can hauntyou as long as you remember it is the past and nothing but. Suicide is a way out, but whatdo you gain. True, some gain notoriety by committing suicide, but you end your life beforeyou have a chance to do anything. Granted, for some people who have nothing going forthem, this is the only way out. Well, then so be it. If you have self-esteem and you listenedto what they taught us in g rade school, you are *censored*. You must remember to never let thatself-esteem turn your head into a balloon. The minute it does, you are gone, and very fewthings can reclaim you. In turn for selflessness, you should find yourself a happy person,and by god, people will like you. Though this can not be attained by reading a book, youcan read about it. Men get away with itbecause they are men.) Pleasure needs to be felt in order for happiness, or so it seems. According to the Hindu religion, pleasure is something fatal. And if you really think aboutit, it is. Lust of chocolate can kill you if you eat to much over a period of time. Lust offood can do the same. While pleasure is not lust, and lust is not pleasure, pleasure is theeffect from lust (usually). Say there is a undercover type person who is in love withsomebody. That person gives them pleasure. If the undercover type person gets into asticky situation, guess what the first pawn to go is- yep, the love interest. Why? Becausethe most pain is mental pain. If you kill the love interest, the undercover person has noreason to live, and will probably do anything to keep that person from being hurt. Italways happens. Mans weak spot is pleasure. Whether it be sex, drugs, or food, they canalways break under the pressure of having it taken away. Man also thinks he wants worldly success. Ever hear that its lonely at the top?Then why the hell would you want worldly success? Worldly success is a facade in whichpeople can take care of low self-esteem. If the world loves you, than nothing can hurtyou. But if you do not love yourself, nobody else can love you. Thats bull because moststars do not love themselves, yet they have girls swarming them. Although I will say thatone night stands are not truly love, but those girls will do anything for it. If you haveworldly success, the world bows down to you and you can do just about whatever youwant and not get caught. The theory is that if you have power, fame, and fortune, you canachieve anything. And thats true because people will do anything if you wave the rightamount of money in their face. But when you go home at the end of the day and you arein your bed all alone, how does one feel? Sure, youll go on, but at the end of your rope,if you have no one to be happy with and arent very happy with yourself, youre going todie unhappy and take all your acquired wealth, fame, and power with you. This is becausehe went after meaningless objects and not after his self. To find out more about yourself,one must look inward, not outward to the cold, harsh reality of the material world. I betyou fall in bed too easily with the beautiful girls who are shyly brave and you sell yourselfas a man to save, but all the money in the world is not enough. I bet youve long sincepassed understanding what it takes to be satisfied youre like a vine that keeps climbinghigher but all the money in the wor ld is not enough. And all the bridges blown away keepfloating up (Liz Phair, 61). Man obtains things on his way through life, whether heuses them or not on his quest for liberation is his choice. Man needs to learn that it is hischoice, and no one elses. Man will go after this until he can run after it no more andeventually he will figure out that each time he finds something it takes more and more tosatisfy his need and hunger for whatever it his he lusts after. Then he will realize that hehas been running in the wrong direction for sixty years, and he will die an unhappy manbecause he cannot change his past. Siddhartha experiences the first of the three things that man thinks he wants inKamalas pleasure garden. He and Kamala screwed each other until the high heavensbrought the monsoons back for their twentieth anniversary. And not once did Kamala getpregnant. No, she got pregnant when Siddhartha left. (How quaint.) Siddhartha andKamala could never really love each other, which I thought was funny. They wereincapable of loving, but in a time period such as the one they lived in, they could get awaywith just screwing each other? Something is screwed up here, no pun intended. Siddhartha had pleasure here because he loved somebody, or at least had a lady friend tospend time with. Early one morning the sun was shinin, he was layin in bed, wondrin ifshed changed at all, if her hair was still red. Her folks they said their lives together surewas gonna be rough. They never did like Mamas homemade dress, Papas bankbookwasnt big enough. And he was standin at the side of the road, rain fallin on his shoes,heading out for the old East Coast, lord knows he paid some dues, gettin through. Tangled up in blue. She was married when they first met, soon to be divorced. He helpedher out of a jam I guess, but he used a little too much force, and he drove that car as far ashe could, abandoned it out West. And they split up on a dark sad night, both agreeing itwas best. And she turned around to look at him, as he was walkin away and she said?This aint the end. Okay, so the quote doesnt fit Siddhartha and Kamalasrelationship exactly, but the similarities are defiantly there. They met each other one night,had a pleasurable twenty years together and then ended up splitting up because Siddharthahad to go find himself. Siddhartha also has pleasure in working with the Kamaswami and being amerchant. Siddhartha becomes rich working with the Kamaswami, and obtains many finethings: clothes, food, wine, etc. Siddhartha can gamble his money and binge drink fordays, just dwindling his money away, but he gains it back somehow. Siddhartha gainsfame and wealth here, and therefore attains power. He can do anything he wants, when hewants, where he wants and the townspeople will go along with it because he has money inthe bank and they dont. Siddhartha enjoys great pleasure with this, and ends up hatinghimself for it in the end. But he needed the experience of what it is like to be filthy rich inorder to attain Nirvana at the end of the novel. Siddhartha experiences wanting everything in the world throughout his time in thevillage where Kamala lives. This is due to him being of mind and state to be able to attainalmost anything he wants. He can ask ask and ask and ask for things, and most of themwill come to him because he has wealth and fame and power. He can satisfy his needs formaterial objects because he has money. He can gamble all his money away because therewill always be more money that will come into his hands. More money that he can dowhatever he wishes to do with it. Siddhartha has wealth and fame and power he and canattain whatever he wants except for the fact that hes out to attain Nirvana and he will notattain it by wanting material items. Having money and nice possessions is nice, but heneeds to become one with god, and god has no possessions. God is everything. Siddhartha realizes that his relationship with Kamala was good and evil, and theymeet again when she comes to the river to go on her pilgrimage to the Buddha. Shebrings with her Siddhartha, her son. Siddhartha tries to love his son, but his son will notlove him back, and this is the final thing that Siddhartha must realize: his son will neverlove him, as he will never love his father. Siddhartha realizes what his life is for andrealizes what he met Kamala for. He had a job in the great north woods, working as acook for a spell. But he never did like it all that much and one day the ax just fell. So hedrifted down to LA, where he reckoned he tried his luck, workin for a-while in an airplaneplant loading cargo onto a truck. But all the while he was alone the past was close behind. He seen a lot of women but she never escaped his mind and he just grew tangled up inblue. She was workin in a topless place and I stopped in for a beer. I just kept lookin atthe side of her face in a spotlight so clear. And later on as the crowd thinned out, Is aboutto do the same. She was standing there in back of my chair, sayin to me ?Whats yourname? I muttered something underneath my breath, she studied the lines on my face. Imust admit I felt a little uneasy when she bent down to tie the laces of my shoes. Tangledup in blue. She lit a burner on the stove and offered me a pipe. ?Thought youd never sayhello, she said, ?You look like the silent type. And she opened up a book of poems andhanded it to me, written by an Italian poet from the thirteenth century. And every one ofthem words rang true. And glowed like burning coal, pouring off of every page like it waswritten in my soul from me to you, tangled up in blue. He was always in a hurry; too busyor too stoned. And everything she ever planned just had to be postponed. She thoughtthey were successful; he thought they were blessed with objects and material things, but Inever was impressed. And when it all came crashing down I became withdrawn. The onlything I knew how to do was to keep on keeping on like a bird that flew. Tangled up inblue. So now Im going back again, got to get to her somehow. All the people we used toknow, theyre an illusion to me now. Some are mathematicians, some are doctor wives. Dont know how it all got started. Dont know what theyre doin with their lives. But me,Im still on the road, heading for another joint. We always did feel the same. We juststarted from a different point of view. Tangled up in blue (Bob Dylan, Tangled Up InBlue). Siddhartha leaves the path of desire when he leaves the village and goes walking. Siddhartha decides that to become one with god and Atman-Brahmin, he must leave thevillage after twenty years of dealing in the dirty material world. Siddhartha decides to gosit under a tree and think about what his life means to him- about what his life is for. Hefalls asleep (much like the real Buddha, also named Siddhartha) under a tree and realizesall he has just done. Since you must experience bad things to attain Nirvana, he did good,but his soul is tarnished and he must cleanse it somehow. His old friend Govinda comesalong and they speak for a while about what theyve been doing with their lives. Siddhartha is disillusioned and must find a way to cleanse his soul, so he goes walkingfurther and he finds the ferryman, Vesudeva. Man really wants liberation because man must be alone to be truly at peace. Otherpeople screw up your karma by giving you negative vibes. Think about it logically, if youare by a person you do not like all that much, what do you do? Act like you love them?No, generally you can feel the tension because you dont like being around that person. Thats what bad vibes are, the tension you feel around something you dont like. Yourkarma does not need this tension, it needs to feel loved. To attain Nirvana, you must loveeverything to become one with everything. You must understand that to be one witheverything is to love yourself because you are the universe and the universe is you. Therefore if you do not love that person, you are feeling that tension because your karmais trying to love that person. Your karma wants you to attain Nirvana, it doesnt want youto be an amoeba in your next life because, lets face it, being an amoeba isnt all that muchfun. Siddhartha comes to the realization that he needs liberation when he is under thetree. He decides that the way to self-enlightenment is through going inside ones self andfinding god. Finding everything that has ever harmed you and laughing at it. Becomingone with evil and good at the same time, and thusforth liberating ones body and mind. Siddhartha does this by listening to the river for eleven years. He listens carefully toeverything, and takes people back and forth on the river all day. He works in the field forhis and Vesudevas food some days. He becomes a good humble man because he islistening to the river, and the river is telling him that all things are one. Like the river andtime, everything is one in itself because everything is of god and god is of everything. How can one come about Nirvana? By realizing that while a river is a river, it is at allplaces at one time. Beginning, middle, and end, all at the same time, what a concept. Theconcept that Siddhartha understands and learns from when his son wretches his heart fromhim. Siddhartha must let go and let other people find their paths. When the intensity of yellow colour increases, the rate of enzymatic activity also increases. Hypothesis: a) IPTG activates beta-galactosidase enzyme at most effective effects. b) The rate of beta-galactosidase enzymatic activities depend on the time of induction. Materials and Methods: Part A: Time course of induction of ÃŽÂ ²-galactosidase by IPTG Induction of the ÃŽÂ ²-galactosidase enzyme. Two different sets of culture condition were investigated. One set in the condition with IPTG (5mM) and another set in the condition of adding water as the control experiment. 15 labeled microfuge tubes which contain 100  µl of the CTAB solution which used to kills the E. coli cells and lyses the cells to release the contents including ÃŽÂ ² -galactosidase were prepared and placed in the ice bath. 2.5ml of actively growing Escherichia Coli K12 was transferred into two separate 50ml conical flasks and covered with the foil immediately and then immersed in the temperature of 37  °C shaking water bath. 250 ÃŽÂ ¼l of water was added into the control flask and note the time as t=0 and then transferred 200 ÃŽÂ ¼l of the E. coli culture out immediately into the microfuge tube which labelled as 0c tube, mixed well and stored in the ice bath. The same procedure for another set of conical flask but 250 ÃŽÂ ¼l of IPTG was used inst ead of water. After that, two conical flasks were placed in the shaking water bath to maintain the temperature constant at 37 °C.The previous two steps were repeated for preparing the 1, 2, 3, 4, 5, 7, 10, 12, 15, 30 and 45 minute time points for the induction flask and 15 and 45 minutes time points for control flask. ÃŽÂ ²-galactosidase activity of each sample was ready to be observed after addition ONPG and Na2CO3 which used to stop the assay activity by changing the pH value to 11. (School of Biotechnology and Biomolecular Sciences, 2012). ÃŽÂ ²-galactosidase Assay. 15 sample of microfuge tubes were placed in the 37 °C water bath for 5 minutes to reach thermal equilibrium. Addition of 200 ÃŽÂ ¼l of 3mM ONPG into each sample at every 30 intervals and addition of 300 ÃŽÂ ¼l of 1M Na2CO3 into the microfuge tube followed by order after exactly 5 minutes of time of ONPG induction to deactivate the ÃŽÂ ²-galactosidase enzyme activities. The time of ONPG induction was recorded. All samples were centrifuged for 5 minutes and then 300 ÃŽÂ ¼l of supernatant of each sample was taken out and read the absorbance under 414nm with 300 ÃŽÂ ¼l water used as the blank. E. 250 ÃŽÂ ¼l IPTG (5 mM) then, after the 10 min sample is removed, add 250 ÃŽÂ ¼l  chloramphenicol (200ÃŽÂ ¼g/ml). F. 250 ÃŽÂ ¼l IPTG (5 mM) then, immediately after the 10 min. sample is removed,  add 250 ÃŽÂ ¼l rifampicin (250 ÃŽÂ ¼g/ml). G. 250 ÃŽÂ ¼l IPTG (5 mM) then, immediately after the 10 min. sample is removed,  add 250 ÃŽÂ ¼l streptomycin (500 ÃŽÂ ¼g/ml) . Note: At zero time point, all materials were added into set A, B, C and D while the antibiotic that used in the set E, F and G was added after 10 minutes time points sample has been taken out. Discussions: From the graph shown in the figure 2, it can clearly see that the units of beta-galactosidase per ml of bacterial culture show a positive results when IPTG used in the induction but no response when water used instead of IPTG. The longer the IPTG induction time, the greater the units of beta-galactosidase per ml of bacterial culture produced. It can be explained that, IPTG acts as the inducer which depressed the repressor protein into inactive form by undergoes conformational change in the shape of the repressor protein that prevent them from binding to the operator region. Thus, the RNA polymerases can bind to the promoter site without any obstacles, transcription of lac operon occurs. Therefore, it can be concluded that inducer is playing a significant role in inducing of beta-galactosidase enzyme. Model data provided by the coordinator was used instead of the raw data because there is induction timing error when transferred the sample which causes the failure of the group results as it can notice that there is a sudden decrease in the value of the beta-galactosidase per ml of bacterial culture produced during 12 minutes in the figure 1. Based on the information provided in the part B experiment, the highest value of unit of beta-galactosidase produced was observed when IPTG was presence in the culture environment. An increasing trend of response with lower efficiency of effect were shown in the lactose, IPTG (5mM)+glucose and IPTG (10mM)+ glucose induction. However, as rifampicin, streptomycin and chloramphenicol added into the culture samples, a increasing concentration of beta-galactosidase enzyme at the beginning of experiment until 10 minutes then the reactions started to maintain at the constant level. The observations can be elucidated that when both IPTG and lactose were used as the inducer, they play the similar mechanism but the only reason that causes lactose had lower performance is IPTG will not be broken down during reaction whereas lactose will be degraded or used by the cells. As the rate of lactose degradation increases, the concentration of inducer in the culture decreases. In addition, glucose and galactose were formed after hydrolysis of lactose molecule. Glucose molecule is more preferred than the lactose molecule by the E.coli. Hence, a lower performance was shown when lactose was used as the inducer and IPTG always the best choice of inducer to use in the experiment. Since the glucose molecules involved in the experiment, a mechanism named as catabolite repression can be used to illustrated the other two IPTG (5mM)+glucose and IPTG (10mM)+ glucose conditions. Catabolite repression is a mechanism that represses the transcription process by introducing glucose molecules into the reaction since E.coli is more preferred glucose than IPTG while IPTG is essential for switching on the reaction. When the concentration of glucose molecule increases, the level of cyclic-AMP becomes lower. CAMP is required to start the transcription process as it is needed to binds with the Catabolite activator protein (CAP protein) and form an active complex which promotes RNA polymerases binds to promote region. Therefore, if cAMP level is low, there is inactive complex produced it unable to deactivate the repressor protein so repressor protein will bind to o-site and inhibit the occurrence of the transcription process. On the other hand, if the glucose molecule is absence, the high level of cAMP permit the transcription process to take place due to the cAMP binds to the CAP protein to form cAMP.CAP complex and deactivated the repressor protein, transcription takes place. Since the glucose is the preference substrate so when concentration of IPTG increases, it also will not affect the result when glucose is supplied. CTAB solution which also defined as the cetyl trimethyl ammonium bromide and used in the experiment to remove the E.coli cells and also destroyed the membrane of the E.coli cells in order to release ÃŽÂ ²-galactosidase enzyme that needed for the experiment from its content. E. coli is the source of the beta-galactosidase enzyme in the experiment. In the IPTG+ chloramphenicol culture condition, there was only IPTG inside the sample at the first 10 minutes and induction of enzyme was occurs but after Chloramphenicol was added the units of beta-galactosidase of bacterial culture remain constant due to the reason that Chloramphenicol is an antibiotic that inhibit the protein synthesis process and growth of E.coli (Ambrose,P.J,1984). The polypeptides synthesise of the RNA in the E.coli was hindered when added Rifampicin (Campbell, E. A et al, 2001). Rifampicin changes the shape and structure of the ribosomes which makes the lyses of the ribosomes (Sippel Hartmann, 1968) and also preven ts RNA polymerases from binding to promoter region. Executive Summary The gender balance group was given a case study into Tata’s contribution to ethical business practices and sustainability to investigate into. The ten (10) member group shared the research questions amongst themselves for effective and efficient exercise. The group researched into the general view of ethical business practices as behavioral moral values and principles that determine our conducts in the business world. It went deeper to expanciate that, this ethics is applied in various aspect of the business’ â€Å"life†. These include: production process, employees, customers, shareholders, the government and business competitors. The enforcement of this practice goes beyond the provision of code of corporate ethics. Considering both the public and business perspective of sustainability which goes beyond corporate responsibility to the environmental impact of a business’ operations, Life Cycle Assessment (LCA) provides an accurate tool for analyses than carbon footprint of the user phase. This actually takes the true environmental impact of the product from the ‘cradle to the grave’ into consideration. Though this process may be quiet cumbersome, both the business and the environment enjoy a great deal of benefits. Tata contribution towards ethical and sustainable practices is evident in all its activities. It has been able use LCA to produce light metals for the production of cars thereby reducing the amount of CO2 emission. Introduction Steel is a product that is widely used in almost every area of our modern life. It plays a key role in the world’s infrastructure. Tata Steel first established in India is the second largest steel manufacturing industry in the Europe. Its steel products are widely used in the construction industry, aerospace, rail, packaging etc. Tata Steel’s case study into its ethical and sustainability practice was to be addressed by our group – GROUP 27. The group began a series of meetings immediately after its formation. In other to ensure efficient and timely exercise, different tasks were assigned to members and the group leader was unanimously nominated. The questions involved were segmented and given to each member to research on. Data was then collated from various sources and sent the central pull for the preparation of slides and for subsequent presentation on the stipulated date. This report seeks to establish the sources of information for Tata’s ethical and sustainability practices. Key theoretical issues concerning ethical and how a business could use sustainability practice to compete was also given attention to. Since the research was conducted by a group, individuals’ contribution was assessed towards the works completion. Further, my personal observation in relation to the assignment and the knowledge gained were brought to bear. Conclusion from the study was then drawn to evaluate the success or otherwise of the whole exercise. Sources of Information The information gathered was from two main sources – books and the internet. Much information was gathered from Tata’s website which gave me much insight into the research. This actually provided me with Tata’s history, competitors, huge Corporate Social Responsibilities, business ethics and core values among others. Other websites gave me the information about a holistic view of business ethics and Life Cycle Assessment as well. In addition a couple of reading materials including books and journals augmented to the profound knowledge of the general view of ethical behavior and sustainability. Personally I contributed to answering the question of ethical business practices. George Chryssides and John Kaler gave their view as to why businesses should adopt ethical business practices. Materials obtained from the internet suggested that business ethics is applied in all areas of the business including production processes, human resource, shareholders, the community, the government and customers alike. I went further to elaborate on some unethical business practices and what organisations can to do for its enforcement. This gave us much insight to ethical business practices. The outcome was warmly received by members and they profoundly understood the concept and how it was even related to Tata Steel. I was nominated to lead the presentation and the design of the sequence of actions that was to be followed during the presentation was part of my responsibility. On my part, I was to explain what business ethics was, what unethical business practice was and how it could be enforced in an organisation. Theoretical Issues The main objective of the research was to understand business ethical practices, how sustainability could help a business to compete and how Tata Steel had contributed towards such acts. While the objective of any business is to make profits, it should contribute towards the interest of the society by ensuring fair practices. This in effect improve cooperate social responsibility, increase consumer confidence and protect brand values in other to gain competitive advantage. Theoretically, issues concerning ethical business practice and sustainability with particular attention to LCA were of great concern to the research. Ethical business practices are standard behavior of moral values and principles that determine business’ conduct in the business world. Ethical practice takes care of the investor. It is to ensure that the dividends payable to the investor is in safe hands and in good time. Production methods should be environmentally sustainable and everybody involved either directly or indirectly is unduly harmed. Also, it takes into account the health and safety as well as the value of the human capital. Sustainability practice of an organization focuses on the balance between the maximizing shareholders value, honouring the environmental and social responsibilities. For Tata Steel, its sustainability naturally follows its core value of ethics. In all this areas of the challenge, the environmental responsibility happens to be of great concern due to its relationship with the Carbon Footprint. However, findings revealed that LCA provides a more powerful tool to tackling CO2 emission including all phases from production to disposal of material. Even though a holistic LCA is data intensive and time consuming procedure, it enormous benefit ranges from a safer environmental impact, the financial benefit through the knowledge of the resale value at each stage of products life and helps business to gain completive advantage. Research Conclusion From Tata’s case study it was agreed that whereas some businesses do not take ethical issues seriously, Tata steel could be one of the best companies who efficiently and effectively practice business ethics and sustainability. A critical evaluation revealed that the company’s ethical business practice and sustainability is far big thing to write home about. This can be seen in the company’s integrity, understanding of its stakeholders, excellence in business operations, force of unity among employees and corporate responsibility delivered to the society. Personal Observation After researching into the ethical business practices and sustainability using the case study of Tata Steel, some observations were made in relation to the group assignment. Ethical business practices and sustainability has a wider view than I anticipated and has rather broad importance to the business and the society at large. Considering ethical business practices, it was observed that the business has a rather big responsibility to all its stakeholders. It was astonishing to notice that this responsibility is discharged to all those who are directly or indirectly affected by the business. It is somewhat a herculean task for businesses to take care of all its stake holders while maximising shareholders value. However the business enjoys enormous benefits in return of their good deeds. In the light of sustainability, the broader spectrum of LCA was noticed. Though LCA suffers some amount of criticism, a greater number of businesses find it very useful for itself and the environment as well. From Tata’s case study, it became evident that much importance is attached to the whole life cycle than just carbon footprint On a whole, ethical behavior and sustainability in Business operations promote trust and confidence to the consumers and the society in which it operates. These go to promote a good business environment for growth and development opportunities for the company. Knowledge Gained The assignments brought a number of issues to my knowledge. These came from practical activities we undertook and the academic research we embarked on. Practically the effectiveness of division of labour was the first practical knowledge I uncounted. When members were assigned specific task for the presentation, it was surprising to notice that each and every one did what is expected of them making it easier for our preparation. As a team leader, I realised it was always helpful to be a good listener to acknowledge the views of others before making necessary suggestions. This however did not demoralised us from pursuing our course and we were able to complete the task on time. Furthermore, every member of the group had an in-depth knowledge of ethical practice and sustainability from the various discussions held. The Meaning Behind West African Kente Cloth Kente is a brightly colored, banded material and is the most widely known cloth produced in Africa. Although kente cloth is now identified with the Akan people in West Africa, and particularly the Asante Kingdom, the term originates from the neighboring Fante. Kente cloth is closely related to Adinkra cloth, which has symbols stenciled into cloth and is associated with mourning.​ History Kente cloth is made from thin strips about four cm thick woven together on narrow looms - typically by men. The strips are interlaced to form a fabric which is usually worn wrapped around the shoulders and waist like a toga - the garment is also known as kente. Women wear two shorter lengths to form a skirt and bodice. Originally made from white cotton with some indigo patterning, kente cloth evolved when silk arrived with Portuguese traders in the seventeenth century. Fabric samples were pulled apart for the silken thread, which was then woven into the kente cloth. Later, when skeins of silk became available, more sophisticated patterns were created - although the extortionate cost of the silk meant they were only available to Akan royalty.
